There is no Magic Bullet, and i can't show you in one hour or even one day how to lock down your WordPress site. And even then your site can never be completely secure, no more than anything else on the Web.

What we do and How to do it took me over 2 years to learn (on top of years of experience building sites, not learning about them). Everything i know (almost) is at TibetanGeeks.com/technologies/web_development/cms/wordpress/03-wordpress_security/

Why we do it, What blocks us (ourselves), and the Principles behind it, i can speak a little today, to hopefully give some basis for learning more.
